Scientists design compact CRISPR tool to boost genes in mice

Researchers created a small CRISPR delivery system that reduces seizures and strengthens muscles in mouse models of genetic disease.

Hainan Zhang and colleagues designed a single viral package to carry instructions for activating multiple genes within cells simultaneously. This approach solves delivery problems that previously hindered researchers trying to target several genes at once with standard tools.

The new system uses a small protein called Cas12m to cut RNA guides inside cells. Multiple guides share one compact set of instructions for editing genetic targets efficiently. A single viral vector successfully carried eight guides that increased activity in six gene targets for brain and muscle tissue.

In Dravet syndrome models, the treatment raised protein levels and reduced seizures significantly. All treated mice survived the observation period compared to control groups in the study.

Duchenne muscular dystrophy models showed improved grip strength and resistance to fatigue. Benefits persisted six months after intravenous injection in the animals.

Further testing is needed across different disease models and doses to confirm safety before applying these findings to human patients. Both main experiments used very young mice, requiring closer scrutiny of altering several genes activity levels carefully.
